To open those boxes, you need to collect rainbow twists (chains of blockers on the sides). The boxes show you how many blockers you need to collect before you can unlock them.
The blockers that are already on the board are not enough to open the boxes, so you will need to allow the magic mixers to produce more of them. Do not destroy those mixers until you have collected enough rainbow twists!
In the example above, there is one more twist left to collect, and since the last remaining mixer had just produced one, it's safe to set off the combo and clear everything.
